چکیده

We describe a simple, direct, and specific manual method for quantitative determination of total cholesterol in serum. In the method, which requires no extraction of the serum, a single stable reagent and 50 l of sample is used. Serum cholesterol concentrations determined by this method did not differ statistically from those obtained by the reference method of AbeIl et at. Hemoglobin, bilirubin, and y-globulin do not interfere unless present at markedly supranormal concentrations. Inter.run precision is about ±3% (95% confidence limits).

Serum cholesterol assay using a stable Liebermann-Burchard reagent.

A single stable Liebermann-Burchard reagent was prepared and studied for the rapid and accuratedeterminationof total cholesterol in serum. The preparation, stability, and developed spectral character of the proposedreagent were assessed.Reproducibility, recovery, and interference studies on pure standard and patient sera were performed using the Abell-Kendall method as reference. The influence ...

An improved automated determination of serum total cholesterol with a single color reagent.

An anhydrous color reagent containing ferric chloride is pumped through Tygon tubing to a glass coil in a 950 heating bath. Manually prepared serum extracts (1:20 isopropanol dilution) are presented to the stream of preheated color reagent, and the two are passed through three mixing coils, connected in series. The absorbance of the resultant color is determined at 550 m in a 15 mm. tubular flo...

The determination of cholesterol in serum by persulphuric acid oxidation.

A method for the determination of total cholesterol in serum is described. Optimum conditions are found for the isolation of the steroid free from extraneous chromogens, and a new sensitive colour reagent is introduced. The procedure is convenient for routine application, the final colour being light-stable and reasonably time-stable at room temperature. Comparisons are made with one other meth...

Evaluation of the ABA-100 procedure for total and direct bilirubin in serum or plasma.

We present a modified Evelyn-Malloy method, adapted to the Abbott Biochromatic Analyzer (ABA-100) for the determination of both total and direct bilirubin. It requires less than 25 mul of serum or plasma. At least 50 samples can be assayed per hour with economy of reagent consumption and the precision compares favorably with that for conventional manual methods.

Reagent for the enzymatic determination of serum total cholesterol with improved lipolytic efficiency.

We describe a sensitive method for quantifying the extent of cholesterol ester cleavage during enzymatic assay of total cholesterol in serum. Lipids are extracted from the assay mixture with chloroform/methanol (1/1 by vol), concentrated, then quantified by "high-performance" thin-layer chromatography. Although with conventional enzymatic reagents for determination of serum total cholesterol th...
